ToggleThe Clinical Paradigm: An Unmet Medical Need
Conventional cancer treatments—such as systemic chemotherapy and radiotherapy—predominantly eradicate bulk, rapidly proliferating tumor cells. While patients often achieve temporary remission, disease recurrence and distant metastasis remain the leading causes of cancer-related mortality.
At the core of this failure lies a rare, resilient subpopulation representing only 0.01% to 2% of the tumor mass: Cancer Stem Cells (CSCs). CSCs possess inherent survival advantages that allow them to evade standard cytotoxicity, re-seed secondary tumors, and drive therapeutic failure. Understanding and selectively eliminating this elusive subpopulation is paramount to achieving true clinical cures.
Defining Hallmarks of Cancer Stem Cells
Unlike non-stem tumor bulk, CSCs display dynamic plastic states, shared features with adult somatic stem cells, and distinctive oncogenic mechanisms:
- Sustained Self-Renewal & Tumorigenicity: A minute fraction of CSCs can regenerate the entire cellular hierarchy of a tumor, driving clonal evolution and phenotypic heterogeneity.
- Cellular Quiescence & Plasticity: Rather than dividing perpetually, many CSCs reside in a slow-cycling or dormant state (G0 phase), rendering them impervious to antimitotic agents designed to kill rapidly dividing cells.
- Multi-Drug Resistance (MDR) Machineries: CSCs consistently upregulate ATP-binding cassette (ABC) multidrug transporters, enhance intracellular detoxification (e.g., elevated ALDH1 activity), and augment DNA damage response (DDR) pathways to repair therapeutic lesions rapidly.
- Surface Biomarker Signatures: Depending on tumor lineage, CSC populations are commonly characterized by distinct molecular markers, including CD44, CD133, CD24, EPCAM, and active stemness transcription factor networks.
The Intricate Biology of Resistance & Metastasis
CSCs do not operate in isolation. Their persistent survival is orchestrated through multi-layered biological circuits:
- Transcriptional Networks & “Undruggable” Targets: Aberrant stemness programs governed by master transcription factor cascades enforce pluripotency and block terminal differentiation.
- Metabolic Flexibility & Organelle Crosstalk: CSCs dynamically switch between oxidative phosphorylation and glycolysis, relying on metabolic hubs such as glutaminolysis and fatty acid synthesis (FASN) to withstand microenvironmental hypoxia and nutritional stress.
- Niche-Mediated Immune Evasion: CSCs proactively remodel their immediate microenvironment via paracrine signaling and extracellular vesicle trafficking, evading immunosurveillance while priming pre-metastatic niches.
How YAS Lab Tackles the CSC Challenge
Drugging "Undruggable" Stem Cell Regulators
Targeting nuclear transcription factors has historically been considered undruggable. Our lab specializes in identifying transcriptional vulnerabilities within the CSC regulatory core—such as the PBX1 transcriptional axis—and developing novel small-molecule inhibitors to disrupt critical protein-DNA interactions, dismantle the stemness network, and re-sensitize platinum-resistant tumors.
Intercepting Metabolic Plasticity & Organelle Signaling
We investigate the functional crosstalk at organelle junctions—including endoplasmic reticulum-mitochondria contact sites—and unpick the compensatory metabolic circuitry that shields CSCs during targeted treatments. Disrupting these metabolic adaptations deprives CSCs of the energetic plasticity required for post-treatment rebound.
Nanomedicine, Extracellular Vesicles & Novel Therapeutics
Leveraging bio-engineered vesicles, natural immunomodulatory architectures (such as NK cell-derived exosomes), and neoantigen profiling, we engineer next-generation platforms to precisely deliver payloads into the protective CSC niche, bypassing classic multi-drug resistance pumps.
From Bench to Bedside: Our Vision
Eradicating cancer demands moving beyond shrinking bulk tumors to eliminating the stem-like seeds of malignancy. By dissecting the molecular dependencies of CSCs, YAS Lab aims to translate pioneering biological insights into first-in-class targeted therapeutics and durable therapeutic paradigms for patients facing resistant, recurrent disease.
